THE MEASUREMENT OF INSULIN-LIKE GROWTH FACTOR 1 IN SHEEP PLASMA

L.A. Bruce, T. Atkinson, J.S.M. Hutchinson, R.A. Shakespear and J.C. MacRae

A method is described for the radioimmunoassay (RIA) of insulin-like growth factor 1 (IGF-1) in neutralised formic acid-ethanol extracts of sheep plasma. The ability of the acid-ethanol pretreatment to remove the IGF-1 binding proteins (BPs), which interfere in the assay has been examined. Comparative plasma IGF-1 concentrations determined by the method correlated closely (P<0.001) with corresponding values where BPs were removed by acid gel filtration. The method has been applied to studies in which sheep were given exogenous growth hormone and indicated that plasma IGF-1 levels respond rapidly to the onset and termination of treatment.
