DOSE-RELATED PERSISTENT PROLIFEROGENIC ACTION OF CORTICOTROPHIN 1–24 IN NORMAL ADULT HUMAN ADRENOCORTICAL CELLS IN PRIMARY TISSUE CULTURE

A pharmacological dose ofβ-1–24-corticotrophin (ACTH 1-24) induced full differentiation of normal adult human adrenocortical cells in primary tissue culture and even hypertrophy of their subcellular organelles (Armato, Nussdorfer, Andreis, Mazzocchi & Draghi, 1974). It also stimulates synthesis of nucleic acid and mitotic activity (Armato, Andreis, Draghi & Meneghelli, 1975). This communication concerns the effects of a much wider range of ACTH 1–24 concentrations, including physiological concentration, on uptake of [3H] thymidine and induction of proliferation in primary normal adult human adrenocortical cells.

From five consenting adult patients undergoing urological surgery, wedge biopsies of normal adrenals were obtained, decapsulated and cultivated as previously described (Armato, Andreis, Draghi & Nussdorfer, 1974). From day 16 in vitro some of the cultures were treated for either 2 or 7 days with daily doses of ACTH 1-24 differing in order of
